The candidate, during his Ph.D. education, is going to study the changes of autophagic processes in connection with microRNA expression and arrhythmogenesis focusing on ventricular fibrillation in isolated ischemic/reperfused (I/R) myocardium obtained from diabetic and nondiabetic rats under in vitro conditions. Different autophagy-related proteins including LC3II, Beclin-1, Atg-5, Atg-7, and Atg-12 and micro RNA expression will be investigated in spontaneously beating and electrically paced isolated hearts. Based on preliminary results we expect substantial differences in the autophagic processes between the nondiabetic and diabetic myocardium, however, further experiments have to be carried out to precisely understand the mechanisms of arrhythmogenesis in connection with microRNA expression. These experiments are important to understand whether there is or no difference in autophagic processes in nondiabetic and diabetic myocardium in connection with microRNA expression. Cardiac function (heart rate, coronary and aortic flows, left ventricular aortic pressure) and infarct size (TTC staining) will be also registered and measured in isolated rat hearts. Moreover, the incidence of reperfusion-induced ventricular arrhythmias including ventricular tachycardia, ventricular fibrillation will be also detected in nondiabetic and diabetic myocardium.
