Grapevine is a perennial, woody plant, plantations are maintained for decades. If the detection methods are not sensitive enough both the rootstock and the scion, used for their propagation can be infected with different types of viruses In a vineyard virus infection can be transmitted also from the surroundings by viral vectors. Virus infected grapevine itself can survive for a long period of time without symptoms. Specific and cumulative symptoms can emerge if infection with other pathogens (bacteria and fungi) deteriorate the health of the host and if viruses form different families simultaneously are present. Plant viruses have different type of silencing suppressor proteins (VSRs) which are able to block RNAi, the plant defence system, at different points by different mechanisms. If there are several viruses present in the same plant, effects of their VSRs can be added. To analyse this question we should be aware not only of the presenting viruses, but their possible VSRs. Plant viruses are present at a field as viral clouds containing several different varieties of the same virus. These population of viruses can infect different hosts which could lead to their divergent evolution. In the proposed project we would like to investigate the presence of potential VSRs of recently described grapevine viruses. Moreover, we would investigate differences of an already described VSR of Raspberry bushy dwarf virus, which virus can infect not only raspberry but grapevine as well. Our results will identify the presence of possible new VSRs. Moreover it will show how divergent VSRs of the same virus on different host can be. This findings would broaden our knowledge and help to understand not only this aspect of virus evolution, but the fact how multiple infection with different, but latent viruses can culminate in severe symptoms in an important agronomical plant.
