MOLECULAR CHARACTERIZATION OF A NEW NON-CODING RNA (AS-MGMT) IN CONTEXT OF GLIOBLASTOMA MULTIFORME

Description of the research topic:

Glioblastoma multiforme (GBM) is the most common primary tumor of the central nervous system, with a median survival of approximately 15 months from diagnosis. The standard therapy for GBM involves surgical resection followed by radiotherapy and chemotherapy with temozolomide (TMZ), a DNA-alkylation agent that induces DNA breaks and triggers apoptosis of glial cells. Although TMZ significantly increases survival in GBM patients, resistance to TMZ can develop, leading to tumour progression. It is wildly accepted that the expression of the enzyme O6-methylguanine-DNA methyltransferase (MGMT) is the most important factor for TMZ resistance. MGMT is a DNA repair enzyme that protects the cellular genome from the mutagenic effects of TMZ and reverses treatment-induced DNA damage, leading to reduced chemotherapy efficacy. A deeper understanding of the pathogenesis of GBM may reveal additional druggable molecular targets for the development of new therapeutic options by elucidating the genetic and epigenetic factors involved. We focus on the molecular and epigenetic characterization of a newly discovered non-coding RNA in the pathogenesis of GBM. Research work will include cloning, establishing cell lines, investigating cell viability, proliferation rate, invasion capacity, metabolic activity and epigenetic profile analysis.
