Description of the research topic:
The fine-tuning of actin’s functions, thus cellular behavior, involves the concerted actions of actin-associated proteins and their isoforms. Despite its importance, the molecular mechanisms underlying the coupling between actin’s function and the isoform-specific pattern of proteins are unexplored in many cases. The balance of protein isoforms is essential for optimal functioning of the cell, the imbalance often results in pathological conditions and human diseases. Therefore understanding these processes truly at the molecular level is a prerequisite for developing successful preventive and therapeutic strategies.
The research strategy and aims are:
1. to study which isoform-specific characteristics of actin and actin-associated proteins are responsible for the regulation of the morphology, dynamics and molecular composition of cellular actin assemblies;
2. to develop and optimize biomimetic systems reconstituting the isoform-specific functional coupling between actin-associated proteins;
3. to describe the molecular mechanisms underlying the isoform-specific cellular dysfunctions and identify factors that specifically target the protein isoform-specific pathological conditions of the actin cytoskeleton.
The methodology is based on molecular and cell biological tools, protein biochemical and biophysical techniques, fluorescence spectroscopy and microscopy, as well as microfluidics and biomimetic approaches.
