Description of the research topic:

"Reaction of 2-aroylbenzyl chlorides with sodium cyanide afforded, in addition to the expected products, i.e. the corresponding benzyl cyanides, 1-cyano-1-arylisobenzofuranes in comparable yields. 1-Cyanoisobenzofuranes can be considered as versatile starting compounds for the synthesis of drug substances, e.g. via their transformations to the corresponding carboxylic acids or amines. In the course of preliminary studies on the acidic hydrolysis of a 1-cyano-1-arylisobenzofurane derivative, the formation of a surprising dimer-like product was also observed.

The tasks of the PhD candidate will be:
• to continue these preliminary studies and to investigate the synthetic methods in detail,
• to extend the reaction with sodium cyanide to (on both phenyl groups) variously substituted 2-aroylbenzyl halogenides,
• to find the optimal reaction conditions to maximize the yield of the individual products,
• to extend the reaction with sodium cyanide to variously substituted 2-alkanoylbenzyl halogenides,
• to extend the reaction with sodium cyanide to variously substituted 2-formylbenzyl halogenides
• to investigate the acidic hydrolysis of various 1-cyanoisobenzofurane derivatives.

Potentially, a flow chemistry approach can also be implemented and be a part of the dissertation.

High-performance analytical techniques (2D NMR, LC-MS, single-crystal X-ray, etc.) will be used for the characterization of new compounds. The planned synthetic routes and further details can be discussed in a personal conversation. "
