Synthesis and characterization of tissue-specific magnetic resonance imaging (MRI) probes based on manganese(II)

Description of the research topic:

Magnetic resonance imaging (MRI) using gadolinium (Gd)-based contrast agents (GBCAs) is an indispensable diagnostic tool for clinics. However, in 2007 GBCAs were directly linked to the onset of nephrogenic systemic fibrosis (NSF), a rare progressive fibrotic condition that affects multiple organ systems in the tissues of patients with chronic kidney disease or renal failure. Later, in 2014, abnormal signal intensity was reported in non–contrast enhanced T1-weighted MRI of certain brain regions in patients with a prior history of GBCAs use. These serious problems highlighted the flows of GBCAs that were considered to be safe and catalyzed research into paramagnetic contrast agents based on essential metal ions. Mn(II) complexes are undoubtedly the best candidates to to overtake the replace of Gd(III) in CAs research. Over the past 10 years, several ligand platforms were proposed for Mn(II) complexation and were found form thermodynamically stable and inert complexes with Mn(II). In a continuation, the current PhD project aims at the design, synthesis and investigation of macrocyclic ligands derived form pyclen, O-pyclen and bispyclen those Mn(II) complexes can be used as tissue specific MRI probes (liver specific and probes capable of Alzheimer’s disease detection will be synthesized). The PhD student to be recruited will be involved in the synthesis and purification of ligands, the physicochemical (stability of complexes formed with endogenous metal like manganese(II) and Fe(III) ions, inertness of the Mn(II) and Fe(III) complexes) by using pH-potentiometry, UV-visible spectrophotometry, 1H-, 17O-realxometry, etc. analytical methods and structural characterization of complexes (using multinuclear NMR in the solution, single crystal X-ray diffraction studies in solid phase supported by DFT calculations).
