Description of the research topic:

Age-related regulatory alterations may be assumed in the background of middle-aged obesity and aging anorexia since they also appear in other mammals. Our previous studies described such regulatory alterations of leptin and its downstream mediators such as the central catabolic melanocortin system or the central catabolic corticotropin system. Weak catabolic responsiveness to leptin, to alpha-melanocyte stimulating hormone or corticotropin-releasing factor (CRF) in middle-aged and a strong responsiveness in old age-groups. In the PhD program we aim to investigate the effects of long-term (at least 12-week) treadmill or running wheel training on age- and nutritional state-associated alterations in the central catabolic mediator systems (and those of their receptors). We aim to compare the effects of training with those of long-term (at least 12-week) caloric restriction.
Animal experiments will carried out in different age-groups (from juvenile to old) of laboratory rodents. Regarding nutritional state, we establish ad libitum fed and diet-induced obese groups withing the age-groups.
During the analysis of central acute and chronic catabolic mediator responsiveness, food intake is to be recorded in an automated FeedScale system (Columbus), circadian rhythm of core temperature, heart rate, spontaneous locomotor activity in a biotelemetric system (MiniMitter, Respironics). These measurements will be complemented by the registration of oxygen consumption, core and tail skin temperature (the latter indicates heat loss) in an OxyletPro system (Panlab) for indirect calorimetry. The changes in central mediators and their receptors will be investigated with the RNAscope method.
