Description of the research topic:

Sarcomeres are extremely highly ordered macromolecular assemblies where structural organization is intimately linked to the functionality of these contractile units. Although the structural basis of actin and myosin interaction is revealed at a quasi-atomic resolution, much less is known about molecular organization of the I-band, centered around the Z-disk, and the H-zone, located along the M-line. In addition, the mechanisms of thin filament elongation also remained obscured. Thus, acquiring the precise molecular architecture is indispensable in order to understand the details of sarcomere assembly and function in healthy and disease conditions. Our model system, the Drosophila indirect flight muscles are ideal candidates for structural studies, since the IFM sarcomeres have an extremely regular, near-crystalline structure. During our preliminary studies, by using a dSTORM approach, we already determined the distribution of 27 muscle proteins in 1 day old adult muscles, and compiled a refined sarcomere model with a previously unprecedented scope and
resolution. Our current goal is to extend these studies to other developmental stages and describe the structure of the developing muscles in wild type and in appropriately chosen mutant backgrounds. We expect that these experiments will greatly advance our knowledge on the
mechanisms of sarcomere assembly that may help to better understand the different types of myopathies and other muscle diseases.

Practise in Drosophila genetics, molecular biology and confocal microscopy would be an advantage.
