Description of the research topic:

Grape phylloxera, Daktulosphaira vitifolie is a major pest of grapevines. The viticulture industry applies resistant rootstocks as primary form of grape phylloxera management. Our Department is maintaining an internationally recognized grape rootstock variety collection. On the base of the large variability of genetic background of grape rootstocks we are planning to enhance our knowledge if plant secondary metabolites plays important role in susceptibility or versus resistance against grape phylloxera induced leaf gall formation. Insects that manipulate plant development show strategies that remodel the cell content and structure of plant tissues at their feeding site. Number of studies reported reduced plant defensive response close to the feeding sites due to the low level of phenolic compounds. Secondary metabolites, like phenolic compounds are taken into account as host plant defensive and/or parasite inhibiting molecules beside their beneficial gustatory and health properties. As a continuation of our previous study based on the classification of the phylloxera induced differential leaf gall characteristics on 19 different internationally recognized rootstock foliage, we offer a research topic connecting the gall formation ability of phylloxera with the secondary metabolite profile of different infested rootstock leaves. In the frame of the program High Pressure Liquid Chromatography (HPLC) analysis will be applied to identify the up- or down regulated state of secondary metabolites on leaves with Phylloxera induced galls as a part of an established in-vitro culture of 19 internationally recognized rootstocks.
