Acute and chronic pancreatitis (AP and CP) are the two major inflammatory diseases of the pancreas, that can lead the patients to serious, life threatening condition.The mortality rate of severe AP is still unacceptable high (30-50%) whereas CP can turn to pancreatic carcinoma, the most malignant cancer type.Moreover, none of the above mentioned diseases have specific pharmacological treatment.Despite the frightful clinical situation, our current understanding of the initiation, development and progression of AP and CP is largely deficient. We have recently shown that disrupt level and function of cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel plays a crucial role in the early phase of AP and apparent in CP as well (Gastroenterology 2011, Gastroenterology 2015–the latter one is highlighted by Nature Reviews HG). One of the leading histological features of CP and recurrent pancreatitis is the formation of a protein gel in the fine pancreatic ducts, while the main pathological feature of cystic fibrosis is the accumulation of thick and sticky mucus.Therefore understanding the mucus homeostasis of the pancreas is highly important both in physiological and pathophysiological aspects. In this project we will first plan to understand the characteristics of mucin homeostasis in human and after that, we will go to the bench side and will use sophisticated, new generational biomedical techniques to get answers for our questions.The project focuses on cross-functional collaborations between clinicians and researchers.The results of this project may open up the possibility of pharmacological therapy of AP and CP leading to reduced morbidity and mortality
