Selection of hepatoprotective agents in vitro in xenobiotic-induced liver damage

Description of the research topic:

Liver act as a key organ in the metabolism of several xenobiotics. In our work, primary hepatocytes and human hepatocarcinoma cell line, HepG2 will be used in two- and 3-dimensional (2D/3D) cultivation designs to investigate metabolic function of the liver treated with xenobiotics. In the frame of this PhD project special emphasis will be placed on (i) determination of changes in activities for CYP isoenzymes after addition of drug candidates and (ii) study on the effects of trichothecene mycotoxins on hepatocytes after acute and chronic exposure based on liver function assays and morphological examinations. Further goal of this research is to find potential beneficial properties of probiotics and their metabolites, antioxidants in the prevention of xenobiotics-induced liver damage. The experiments will include analysis of cell cytotoxicity and intracellular/extracellular reactive oxygen species to select hepatoprotective agents. Activity measurements of xenobiotic-metabolizing CYP enzymes will be performed to characterize biotransformation routes of the drug candidates and to establish reliable and precise method for prediction of metabolic drug interactions.
