Description of the research topic:

Newer generation dendritic cell (DC) vaccines must build on the knowledge of fine tune modulation of DC development. Our long-term goal is to explore the role of various transcription factors during embryonic stem (ES) cell-derived DC differentiation. We intend to investigate the impacts of several DC specific transcription factors in ES-cell derived DCs and their progenitors. To achieve this we will use inducible murine ES cell lines in which we can tightly control the expression of the selected transgenes. We intend to perform gene expression profiling experiments and carry out chromatin immunoprecipitation followed by massive parallel sequencing to identify the regulatory sites of the selected transcription factors. In addition we will monitor the expression of several DC specific cell surface markers with multicolor flow cytometry and measure the T cell activation and migratory capacity of the engineered immune cells. These studies will accelerate our understanding about a transcription factor directed differentiation of DCs. In addition, our research might help to produce pluripotent stem cell-derived DCs to be applied for immunotherapy.
