Description of the research topic:

Mutations are heritable changes in the DNA sequence of living organisms. They act as a double-edged sword: on one hand their diversity-generating property makes them essential for evolution, on the other hand, they most often cause deleterious effects on phenotype, leading to or increasing the risk of certain diseases in humans. Our workgroup focuses on the genome of Escherichia coli, the laboratory model bacterium. Our aim is to gather knowledge concerning the processes of mutation and mutation- evasion that is sufficient to allow the fine-tuned control of these events. Such control is especially important from the biotechnological point of view, for while directed evolution requires elevated rates of mutation, the stable maintenance of useful strains and genetic constructs necessitates the minimization of further mutational events. Besides point mutations, we also focus on large scale genomic rearrangements caused by the transposition of mobile genetic elements. Our toolbox mainly comprises molecular biology techniques, with special attention to molecular cloning, bacterial genome engineering and transcriptional silencing. As a result of our work, we not only wish to take significant steps in fundamental research, but also to engineer bacterial strains that can be more effectively applied in the overproduction of industrially relevant proteins and high-value compounds.
