Accelerated, random evolution of bacterial genomes in the laboratory

Description of the research topic:

Evolution of genomes is characterized by long, dominant periods of reduction, punctuated by short, rapid expansions. Understanding the driving forces shaping genome complexity is of paramount importance in the era of constructing semi-synthetic microbial genomes. Testing of the hypotheses on genome reduction is hindered by the slow pace of natural evolution, and by the lack of tools speeding up genome evolution in the laboratory. Here we propose a novel scheme of random deletion generation to accelerate laboratory evolution of E. coli, and address the validity of the genome streamlining hypotheses. Applying a novel combination of traditional clone library construction and recombination/selection methods, we plan to generate a large number of random genomic deletions in a cyclic fashion in parallel cell lines, under various environmental conditions. Analysis of the strains will shed light on the genome organization principles, and will produce a blueprint for a minimal E. coli genome.
