Description of the research topic:

The bioavailability of natural food antioxidants is limited because they are present in glycosidic form, and thus, unable to diffuse across the intestinal wall. The dietary intake of these antioxidants can be enhanced through glycosidase-catalyzed deglycosylation of the aglycone. Many zygomycetes fungi can produce exoenzymes in high yield. Besides the hydrolysis, their glycosidases generally display transglycosylation activity as well, thus they can be applicable not only for the liberation of aglycones from their glycosides, but the synthesis of prebiotic oligosaccharides from disaccharides. During our work, zygomycetes glycosidase systems will be elaborated for the liberation of aglycones and/or the synthesis of oligosaccharides. Production of enzymes will be optimized, and analytical and preparative protocols will be applied/developed for the proper identification and purification of the enzymes/products. Analysis of the aglycone/oligossacharide products in food technological model systems is also planned.
