Several lines of evidence indicate that abnormal B-cell functions play a key role in the development of systemic autimmune diseases (SAD), with imbalance in cytokine profile and in natural and pathologic autoantibody levels resulting breakage of tolerance. Our aim in this research project is to find biomarkers and innate signaling pathways which differentially influence the activation and survival of B cell subpopulations resulting disease-specific autoantibody production. We detect the natural and disease-specific autoantibody production of differentially stimulated healthy and pathologic B cells in patients with active systemic autoimmune diseases (SSc and SLE). Regulatory and pro-inflammatory cytokine production and B cell-specific activation markers (SPP1, C3, IFNa) will be detected.
