Analysis of protein O-GlcNAc post-translational modification in malignant hematology disorders

A kutatási téma leírása:

Malignant cells tend to have higher metabolic rate to meet the increased requirements of cell growth and division. Reverting to more primitive, anaerob metabolism is also frequently observed in cancer. The result is a significantly elevated glucose consumption. One of the consequences of increased carbohydrate metabolism is enhanced protein O-N-acetyl-glucosamine (O-GlcNAc) modification. 1-3% of the glucose entering the cells is metabolized through the hexosamine biosynthesis pathway (HBP). The end-product of HBP; UDP-N-acetyl-glucosamine is a direct substrate for a post-translational modification on Ser/Thr moieties. This O-GlcNAc modification is a reversible, dynamic process in the cytoplasm and the nuclei. O-GlcNAc is increasingly recognized as a regulatory mechanism for many intracellular processes including signal transduction, phosphorylation, transcritional activity or protein degradation.
Detecting and analysing O-GlcNAc modification in malignancies has a great potential for the better understanding of the underlying patho-physiological mechanisms. Even more importantly, it might provide a useful diagnostic tool for the detection, risk assessment and monitoring of malignant disorders. Thus our aim is to develop an analytical method applicable for the clinical laboratory and based on their O-GlcNAc levels, to characterize malignant hematologic disorders such as CLL and CML.
