Synthesis and characterization of novel anticancer candidate complexes with improved selectivity based on hypoxia-activation

We have been working for some years on the synthesis, characterization and biotransformation reactions of half-sandwich type Ru(II)- or Os(II)-bioligand (e.g. enzyme inhibitor) complexes with potential anticancer activity. The aim of the present Ph.D. research topic is to study the possibilities for the development of novel dinuclear complexes with potential anticancer activity, incorporating a half-sandwich platinum group metal and an octahedral Co(III) core linked by a functionalized bioligand. Due to hypoxia in cancer cells the reductive milieu is expected to enhance the reduction of the Co(III) and cleavage of the half-sandwich metal-bioligand complex from the parent species is believed to occur. Moreover, careful tailoring the kinetic features of the platinum group metal-bioligand chelate, release of both bioactive entities with potential anticancer activity might also be achieved. As a part of the doctoral work synthesis of Co(III)-(O,O) complexes stabilized with a tetradentate tripodal amine as well as solution equilibrium and redox study on labile Co(II) ternary systems is planned in order to explore the factors affecting the stability of the complexes formed. Another aspect of the work will be the study of the interaction between half-sandwich type platinum metal cores and (N,N) donor chelating ligands. Furthermore, with the appropriate functionalization of the above bioligands the synthesis and characterization of novel ambidentate ligands with (N,N) and (O,O) donor sets together with their bimetallic complexes as potentially hypoxia-activated compounds with dual anticancer activity are also planned.
The aim of this Ph.D. proposal is to reveal the parameters influencing the outlined metal ion–ligand interactions in order to be able to synthesize the functionalized bioligand derivatives and the above bimetallic complexes.
The successful candidate is expected to carry out the synthesis and characterization of the ambidentate ligands and their mono- and heterobimetallic complexes. Besides, pH-potentiometry will be used in solution equilibrium studies, while for the structural characterization of the complexes various spectroscopic techniques and cyclic voltammetry will be applied.
