Establishment and utilization of genome editing in barley and other crop species

A kutatási téma leírása:

Genome editing is a powerful molecular biology technology introducing site specific mutations into the genome of plants, animals and microbes. The successful routine utilization of genome editing in the laboratory allows the precise investigation of target genes providing an efficient tool to understand complex biological processes, not only in model plants but also in important crop species. We are carrying out experiments for utilizing barley genome editing in our laboratory and we would like to extend these experiments for other important crop species such as various wheat cultivars. By comparative bioinformatics approaches we will identify barley genes involved in RNA interference and in determining economically important traits, such as seed number and/or size, to enhance the production rate of important barley cultivars. We also intend to investigate the developmental functions of RNAi associated small regulatory RNAs influencing the beneficial traits of crop plants. The identified genes will be mutated by genome editing and mutant plants will be investigated under various conditions to reveal the role of candidate genes. The planned experiments will be carried out in Genetic and Biotechnology Institute of MATE which possesses all the equipment and basic instrumentations necessary to carry out the planned experiments at the level of international standards. The successful applicant will carry out standard molecular biology methods (RNA, DNA extraction, molecular cloning, PCR, Q-PCR reactions, Northern and western blotting, plant phenotyping etc.) and basic informatics analyses (Blast search, high throughput sequencing data analyses).
