A kutatási téma leírása:
The complement system plays a crucial role in the innate and adaptive immunity leading to the elimination of invading pathogens. It has been revealed only in the last decade that components of the complement system exhibit non-immune functions in the central nervous system. The classical pathway of the complement cascade is implicated in the selective recognition of synapses leading to their subsequent pruning by microglial phagocytosis. Complement-mediated synapse elimination is obligatory in brain development and in maintaining synaptic plasticity. However, its pathological function leads to aberrant synapse loss in in aging and in neurodegenerative diseases, such as Alzheimer’s disease. Our hypothesis is that well defined synaptic molecular deficits lead to complement tagging and concomitant selective phagocytosis of the labelled synapse. The aim of the research group is to discover the molecular background and mechanism of complement mediated synapse elimination and find potential components that can be targeted to prevent pathological synapse loss. The PhD candidate should participate in this work and investigate the role of various complement components by using proteomical and molecular biology techniques.
