We have already prepared IKP FXIII-B isolated from the plasma of homozygous patients and these patients expressed willingness to donate more plasma if required. We will test such IKP FXIII-B in different experimental set-ups and compare their behavior with the wild type protein. Its binding affinity to FXIII-A and to fibrinogen variants will be measured. It will also be tested if the decreased plasma FXIII level of healthy IKP carriers is related to the accelerated decay of the protein in the circulation. For this purpose FXIII-B KO mice will be used. FXIII-B prepared from wild type and IKP homozygous individuals will be administered to the mice and the kinetic of elimination will be followed by daily measurement of FXIII antigen in the plasma of the animals, using our supersensitive chemiluminescent FXIII antigen assay. Another approach will be the determination of intron K polymorphism of FXIII deficient patient being on prophylaxis with recombinant FXIII-A2 and monitoring the elimination of the recombinant protein in patients with various intron K genotypes.
