Heart failure is a leading cause of death in the developed countries. During the development of the two major heart failure phenotypes, systolic and diastolic heart failure distinct, characteristic changes occur in the shape and function of the left ventricle (LV) (eccentric and concentric LV remodelling). Alterations of cardiomyocyte structure and function play an important role in this process. The main objective of the project is therefore to investigate how oxidative modifications of cardiomyocyte sarcomeric proteins contribute to LV systolic and diastolic dysfunction. During the planned research we aim 1. to investigate the effects of oxidants, oxidative enzyme systems and antioxidants on the contractile function of permeabilized cardiomyocytes derived from human LV myocardium (calcium (Ca)-dependent active force, Ca-independent passive force component, Ca-sensitivity); 2. to explore the effects of post-ischemic myocardial remodelling on cardiomycyte function and on myofilament proteins (sulfhydryl-group oxidation, carbonyl-group formation in an in vivo mouse model of myocardial infarction); 3. to identify alterations in cardiomyocyte mechanics and myofilamentary proteins related systolic and diastolic heart failure. The results of the planned research could largely contribute to our understanding on the relationships between cardiomyocyte dysfunction and posttranslational, especially oxidative myofilament protein modifications occurring during LV remodelling in heart failure
