Description of the research topic:

"ABC transporters are membrane proteins that mediate the transport of various molecules across biological membranes, thus playing a pivotal role in the trafficking of substances across physiological barriers composed of polarized epithelial or endothelial cells.
including the plasma membrane of polarized cells that form the physiological barriers of our body. Thus, correct cellular localization to the proper membrane compartment is crucial for their function. Several mutations in ABC transporters affect the intracellular trafficking rather than the function of the protein. The main objective of the program is to gain a deeper understanding of the cellular routing of wild-type and disease-causing mutants of ABC transporter proteins, with special focus on ABCG2, whose mutations can cause hyperuricemia, gout, or even drug-related toxic reactions. The applied methods include advanced cell culturing, basic biochemical techniques, immunofluorescence staining, and advanced microscopy, such as confocal microscopy and high content screening. The program is not only interesting from a basic science aspect but also holds the potential for practical utilization, since identification of trafficking defects of disease-causing variants helps to identify new therapeutic targets for the treatment or prevention of the ABC transporter-related medical conditions.
