Description of the research topic:

The results of in vitro fertilization (IVF) are still at about 30% worldwide. The success of the procedure depends primarily on two factors: i/ the viability of the embryos to be transferred, ii/ the maternal part (responsiveness). There are many investigations worldwide on the two mentioned factors but unfortunately, at present there are no reliable biomarkers which could predict the efficiency of IVF intervention. The present research would like to analyze both the embryonal and maternal factors as well. For the assessment of embryonal quality, the alkaline phosphatase activity, nucleic acid content and other metabolites (e.g. glucose consumption) are planned to be investigated from spent embryonal culture media. Both from the culture media and from the maternal samples (serum and follicular fluid) we plan to isolate exosomes and measure their size distribution, nucleic acid content and surface marker proteins the data of which would be compared with the success of IVF. We would like to work out a biomarker combination that has a predictive value regarding the IVF procedure.
