Description of the research topic:
In muscle fibers the atrophy/hypertrophy is determined by the balance of protein degradation and synthesis. The common basis of regulation is the insuline growth factor (IGF) receptor and the associated proteine kinase B (AKT). In active, phosphorilated form AKT inhibits the protein degradation via the inhibition of the ubiquitin-proteosome system (E3 ligases, atrogin-1). In the other patway AKT stimulates muscle fiber hypertrophy through mTOR activation. Investigation of these mechanisms in muscular dystrophies and acquired myopathies (eg. inactivity, steroid myopathy) is possible by histology (immunohistochermistry, in situ hibridisation) and molecular biology methods (mRNS measurement by RT-PCR). The degree of regeneration can be specified on histology samples (eg. satellite cell density). Ther results of above tests might be compared to clinical data, perhaps biochemical markers obtained from blood
