Identification of resistance genes against major pathogens and their utilization in potato breeding

Description of the research topic:

Potato is the world's third most important staple food crop, produced from the tropical areas up to the arctic regions. The most obvious protection of potato from the many pathogens it has is the introgression of resistance genes into the new cultivars. Phytophthora infestans, causing late blight and the PVY virus are the most dangerous pathogens of potatoes. The present research program aims to explore the biotic stress response processes and identify late blight and PVY resistance genes. For this end, the whole genome sequence of the tetraploid potato cultivar White Lady will be analyzed for resistance gene candidates, and transcriptomic studies on several other genotypes with known resistance will be performed to understand the infection-induced expressional changes in the genome and for identification of potential resistance genes. Micro- and small RNA-s will also be analyzed to identify their possible role in resistance. The real function of the resistance gene candidates will be analyzed by genome editing, utilizing the CRISPR/Cas9 approach. Agrobacterium-mediated transient expression will be used, and real transformants will be produced. An easily applicable molecular selection system will be developed based on the results.
