Description of the research topic:
Epigenetic modifications are important regulators in plant development, which involves histone post-translational modifications and DNA methylation which are transmitted through DNA replication and cell propagation, thereby determining and maintaining cell type specific gene expression patterns. Histone modification has important role in chromatin remodelling and gene expression. Among the histone modifications, acetylation/deacetylation is a key switch for regulating genome activity such as gene transcription. To study the effect of epigenetic master regulators on tomato leaf development and trichome formation we prepared SlIDM1 histone acetyltransferase mutant tomato lines by genome editing. SlIDM1 is the orthologue of Arabidopsis AtIDM1 that regulates AtROS1 mediated DNA cytosine demethylation. Because SlIDM1 is an orthologue of Arabidopsis AtIDM1 and histone acetylation is usually associated with gene activation, we hypothesize that SlIDM1 might regulate the expression of trichome initiation and leaf development genes by preventing their hypermethylation. During the PhD project we would like to train the applicant to identify the target loci of SlIDM1 in genome wide level. We also plan to characterize the SlIDM1 protein temporal and spatial expression pattern during plant development during the PhD program.
