Description of the research topic:

"We are aimed to identify the post-translational effects of the NO signal on effector elements of plastidial Fe and Mn homeostasis using bioinformatics and proteomic methods. We apply Arabidopsis thaliana lines affected by NO synthesis and S-nitrosoglutathione reductase. Transcriptome sequencing and qRT-PCR validation are performed to analyse the involvement of Fe and Mn homeostasis elements. We plan to investigate the presence and amount of NO using confocal fluorescence microscopy and electron paramagnetic resonance spectroscopy performed on isolated plastics. In a synchrotron instrument centre, we investigate the localization of metals at the cellular level using low-energy, microscopic resolution X-ray fluorescence imaging.
Mössbauer spectroscopy is used to detect ferritin-bound iron. Tyr-nitration and Cys-nitrosylation will be investigated by western blot and biotin switch. Bioinformatics modelling is performed to predict the nitration and nitrosylation sites of elements of plastidial Fe and Mn homeostasis."
