Description of the research topic:
"The larval salivary gland of the fruit fly (Drosophila melanogaster) is totally degraded during development contributing to the material of adult tissues. During regulated cell death in Drosophila salivary gland cells, there is an extensive reorganization of the cell's organelles, including the endoplasmic reticulum (ER). We were the first to observe that this restructuring leads to the fragmentation and vesiculation of the ER. The vesiculation of the ER during Drosophila salivary gland cell death is a developmentally regulated process that involves the fragmentation of the ER and its subsequent degradation by the lysosomes. The molecular mechanisms underlying the ER vesiculation during cell death is not well understood. The specific aims of the doctoral work are:
(1) To clarify whether the ER stress is the cause or consequence of apoptosis activation in pupal salivary glands and check at what extent autophagy takes part in the death process;
(2) to identify signaling pathways involved in the outbreak of ER stress and to find inhibitors to block its manifestation.
The candidate have to take care of the mutant and transgenic fly stocks, to make crossings, isolate salivary gland samples and perform PCR, Western blot, light and electron microscopical experiments and immunocytochemistry.
