In vitro examination of the damaging effects of mycotoxin combinations

Description of the research topic:

Due to the co-occurence of mycotoxins in feeds, determination of their combined toxicological effects such as hepatic and intestinal damage is of key importance. IPEC-J2 cell line as intestinal epithelial cell model and primary hepatocytes will be used in our experiments for examination of interaction among intestinal epithelium/liver and mycotoxins. In the frame of this PhD project special emphasis will be placed on (i) study on the cytotoxic effects of mycotoxins on hepatocytes and on enterocytes after acute and chronic exposure and (ii) determination of changes in activities for CYP isoenzymes after addition of mycotoxin combinations (iii) searching for potential beneficial properties of probiotics and their metabolites, antioxidants in the prevention of mycotoxin combination- induced cell damage.
The experiments will include analysis of cell cytotoxicity and intracellular/extracellular reactive oxygen species in addition to activity measurements of different CYP isoenzymes to characterize the effects of combined mycotoxins and to establish reliable and precise method for prediction of drug bioavailability after mycotoxin contaminations.
