Description of the research topic:

Symbiodinium is a dinoflagellate alga, which is a photosynthetic partner in coral symbiosis. This alga is a very important research target due to the coral bleaching phenomenon, which occurs recently as a consequence of global warming. Despite its importance in aquatic ecosystems, several physiological mechanisms of this algae remained uncharacterized so far, because of the lack of efficient transformation methods. In the project we aim to develop a transformation system for the nuclear genes of Symbiodinium, by using a microfluidic approach to create protoplasts from Symbiodinium cells, which are able to take up oligonucleotides that can be used for the transformation of nuclear genes. With this unique approach, resulting in loss of function of targeted genes and processes we aim to characterize photosynthetic processes in Symbiodinium on single cell level. With this approach we aim to establish a single cell phenotyping system of Symbiodinium sp. in order to gain more insights into the photoprotective mechanisms that help to avoid coral bleaching.
