Description of the research topic:

The skin is our largest organ and the first protective barrier of the human body. The skin is able to resist the invasion of harmful substances to the body and prevent the loss of moisture and electrolytes, provided its barrier is intact. The surface of the skin is covered by a lipid layer, which is mostly synthesized by the sebocytes of the sebaceous glands. The sebaceous lipids (or sebum) represent the majority of skin surface lipids that are essential in the maintenance of the epidermal barrier. Altered sebum production is a major feature in the pathophysiology of

several inflammatory skin diseases, but the metabolic pathways regulating sebum composition and secretion are not well understood.

Growing body of evidence demonstrates that poly(ADP-ribose) polymerase (PARP)2 shapes the lipid metabolic profile in various tissues, and our preliminary findings revealed that PARP2 controls the steroid biosynthesis of epidermal keratinocytes, however, the function of PARP2 in the sebaceous glands is not characterized. We hypothesize that PARP2 regulates both the quality and quantity of sebum, and that the dysregulation of PARP2 in the sebaceous glands is a potential pathogenic factor in the development of skin diseases. Our goal is to understand the role of PARP2 in sebum synthesis in both in vitro and in vivo models, that may contribute to a better understanding of the pathogenic factors of skin disorders associated with altered sebum production.
