Obesity and comorbidities are among the world’s major health and socio-economic problems. Hungary has the highest obesity rate in Europe and 4th among the OECG countries. Maintaining a healthy lifestyle (including diet and exercise) is vital in the prevention of these disorders. Recent studies have revealed a strong association between health and the microbiome in various organisms. The relationship between obesity and the microbiome has been investigated in mouse models. The aim this project is to examine the effect of two dietary methods [low-carb and personalized carbohydrate (CH) intake] and exercise on the microbial community, and on the expression of human and microbial genes. We plan to carry out our analysis based on feces samples of obese and lean insulin-resistant or diabetic participants using cutting-edge genome analysis technologies. Interactions within the microbes and with the host will be examined in vitro. Dogs (puppies and adults) will be used as a model for examining the effect of feeding with varying CH content on the microbiome and on the host cells, along with microbiome inheritance. We expect to obtain useful information about the effects of lifestyle on the microbiome, which in turn, will exert a beneficiary effect on health. On the basis of obtained data, we can suggest dietary strategies and potential clinical therapies (fecal transplantation or pro-biotic therapy) for losing weight and for the treatment or prevention of obesity-associated disorders.
