Description of the research topic:

Detailed examination of key proteins and protein interactions of the postsynaptic density (PSD), which regulate the function and signaling mechanisms of excitatory synapses, will be accomplished during the PhD work above. The corresponding domains and segments of the proteins will be produced in a bacterial expression system. Their function and interactions will be analyzed by various spectroscopic and biophysical methods. Beside these protein interaction technics (BLI, ITC, Fluorescence Polarization), performing NMR spectroscopic measurements is also possible, with which the structural dynamics of the disordered regions can be examined in more depth. Based on the experimental results, we are going to establish structural and dynamic models of the interactions using bioinformatics methods, by which we can even discover new targets for the pharmacological treatment of various neurological disorders such as autism spectrum disorder, schizophrenia, or drug addiction.
