Description of the research topic:
In recent days one of the most extensively developing research field is the developmental biology and stem cells research, which uses the methods and results of anatomy, cytology, molecular biology and genetics. The aim of our study is to investigate the organogenesis of the lympho-myeloid organs, the formation of the hematopoietic microenvironment, which consists of distinct combination/network of stromal cells and their product, extracellular matrix. Accessory dendritic cells are the main candidates in the formation of organ specific environment (bursa Fabricii, thymus, T and B dependent areas of peripheral lymphoid organs). The methods, used in our work, combine the techniques of classical embryomanipulations (chimerism, ablation, tissue recombination, immunocytochemistry, in situ hybridization) with gene manipulation techniques like, retroviral vectors. The knowledge of the microenvironment of the “niche” gives a possibility to produce an in vitro model to study the differentiation and maturation processes of hemopoietic stem cells.
