Description of the research topic:

The mechanism how bacteria regulate their cooperative activities by releasing, sensing and responding to signalling molecules or autoinducers is called quorum sensing (QS). The QS makes possible for a population to behave as a multi-cellular organism in host colonization, biofilm formation, production of virulence factors and adaptation to changing environment. Quorum sensing is a less known mode of communication among fungi; the existence of fungal QS was revealed a few years ago. The direct modulation of fungal communication is a new-wave strategy in order to interrupt cell-to-cell communication necessary for adaptation of microorganisms to their environment. The main objective of the PhD research is to find efficient and natural quorum quenching agents against fungi as well as to explore and describe the underlying mechanisms of QS inhibition (QSI) in different fungal species. Various QS inhibitors will be tested in order to find the feasible agent hindering the cooperative action of fungi. Cyclodextrin-mediated quorum quenching as a novel approach will be also studied. Both free cyclodextrins and further QS inhibitors complexed with cyclodextrins will be investigated. Complex microbiological methodology complemented with chemical analysis of signalling molecules will be applied in order to find the feasible cyclodextrin. Antifungal and anti-quorum sensing activities of QSIs will be determined and compared based on the results of specific microbiological models.
