Description of the research topic:

The PhD research focuses on development of in vitro permeability models using for lead optimization, identification of tissue specific agents or selection of dosage form. The 96-well sandwich plate-based PAMPA (parallel artificial membrane permeability assay) method are able to model drug transport across different physiological barrier, such as gastrointestinal lumen, blood-brain barrier, corneal epithelium, etc. The optimized tissue specific PAMPA models can provide high-throughput, cost effective tools for lead optimization and development of drug formulations. The optimization opportunities of PAMPA models (such as artificial tissue-specific membranes, incubation time and temperature, pH and electrolyte concentration of donor and acceptor sides of plate wells etc.) will be investigated. The concentration gradient between donor and acceptor sides of PAMPA system will be followed by LC-DAD-(MS) or UV-spectrophotometer plate reader. The results of these research can support the different stage of drug research from discovery to dosage form selection.
