Description of the research topic:

The aim of the planned research work is the identification of genes and metabolites having major role in the redox control of the transitions between growth, development, quiescent period and death in callus cultures, root and shoot primordia induced from embrygenic callus or appearing during germination and in shoot and root tips of young seedlings of cereals (wheat, maize). The effect of genetically determined and induced (by direct and indirect oxidative stress) changes in the redox environment on these transitions will be studied. The related genes will be selected for checking their regulatory role during the transitions based on the results of our previous transcriptome analyses in which redox-responsive mRNAs and miRNAs were identified. The related metabolites will be determined in the above described experimental systems by metabolite profiling. The redox environment (reactive oxygen species, antioxidants) and hormone levels will be monitored to see the cross-talks. The expression of the genes having major effect on the transitions and subcellular distribution of the glutathione will be analysed in apical meristems using in situ techniques.
