Description of the research topic:

Anomeric spirocycles are compounds in which the anomeric carbon of a sugar ring is part of another (hetero)cycle. Such compounds can be natural or synthetic products with various biological effects. Thus, the natural papulacandins have antifungal effect, the hydantocidin is a herbicide, while the synthetic tofogliflozin is the active ingredient of recently approved antidiabetic drugs. In our previous research we have synthesized several glucopyranosylidene-spiro-heterocycles which were shown to have glycogen phosphorylase inhibitory effects and diminished blood sugar levels in vivo.
Recently, in collaboration with Swedish partners, some xylopyranosylidene-spiro-heterocycles were shown to inhibit the β4GalT7 galactosyl transferase enzyme that starts the glycosaminoglycan (GAG) biosynthesis. In the present research new glycopyranosylidene-spiro-heterocycles of D-xylo and L-arabino configuration will be designed and synthesized and further tested for the above enzyme in order to better understand the structure-activity relationships.
The Helicobacter pylori bacterium can be found in approx. the half of the world’s population, and is one of the most frequent pathogen responsible, among others, for the formation of gastric ulcers and tumours. Due to the spreading resistance against antibiotics, new therapeutic possibilities must be explored. In H. pylori, the key enzyme of purine synthesis is the adenylosuccinate synthase (AdSS) that can be inhibited by hydantocidin-5’-phosphate. In collaboration with Polish partners new ribofuranosylidene-spiro-heterocycles will be designed, synthesized and tested as inhibitors of the above enzyme.
