Description of the research topic:

Brain pericytes are mural cells located in the duplication of the basement membrane of microvessels, coming in close contact with cerebral endothelial and glial cells. Although discovered almost 150 years ago, pericytes remain largely uncharacterized cells, heterogenic in their morphology and function, only defined by their localization. Pericytes are multipotent, undifferentiated cells, which form a phenotypic and probably functional continuum with smooth muscle cells to regulate key functions of the neurovascular unit. By using in vitro and in vivo transgenic animal models and advanced microscopy techniques (two-photon and superresolution), we will address heterogeneity of pericytes along the vascular tree and in different regions of the brain. We will identify key elements of pericyte contractility and clarify involvement of pericytes in aging. We will also elucidate how pericytes participate in ischemic stroke-associated reduction in local cerebral blood flow and dysregulation of the blood-brain barrier.
