Description of the research topic:

For fertilizing capability, two probabilities exist: for each spermatozoon, all essential attributes are expressed at the right site and time, and for the total population of sperm, the population contains a given percentage of sperm with full fertilizing potential. The ideal in vitro test for sperm quality would simultaneously quantify the attributes representing different features required for fertility at cellular level.
Spermatozoa can be divided into the following domains: head, midpiece and tail, and into subdomains within these regions. The sperm head contains the chromatin, a unique protamine-DNA complex. Abnormalities of the sperm chromatin structure can cause disturbances in fertility, pronuclear formation, early embryo quality and pregnancy outcome. These abnormalities are classified as “uncompensable”, as subfertile males cannot be brought to normal fertility by increasing the inseminate dosage. Sperm cells with uncompensable defects are able to fertilize oocytes; however, the embryo development may be abnormal.
The main goal of the project is to reveal the dynamic changes in chromatin status in seasonally breeding mammals. Domestic horse would be used as model species. The expected results may help to develop better sperm cryopreservation protocols for endangered wild mammals with seasonal reproduction.
