Description of the research topic:

DNA damages frequently occur in the genome during replication or are caused by DNA damaging agents, which affect genome stability resulting in translocations and often leads to cancer formation. Slower repair speed or improper repair can also increase the probability of chromosome rearrangement by causing genome instability, which is one of the early steps of cancer development. In eukaryotic cells, the repair of broken DNA occurs in the context of highly structured chromatin. Chromatin structure alterations are thus thought to be influenced by not only the efficiency and kinetics of repair but also, importantly, the choice of the repair pathway in the cells. Our primary objective is to understand how the DNA breaks could occur in different chromosomal positions. We also try to address what are the major DNA repair pathways and cellular responses induced by the DNA errors. We plan to identify novel proteins activated by the DNA breaks, being present after the repair has taken place, which could potentially serve as a signal highlighting the damaged and repaired region for further processes.
