Description of the research topic:

The transglutaminase enzyme family contains 9 proteins with various functions. The best-known member of the family is Transglutaminase 2 (TG2), which is a unique multifunctional protein. It has several catalytic activities, takes part in cell adhesion, differentiation, apoptosis, phagocytosis and stabilisation of the extracellular matrix. It is involved in the pathomechanism of tumour autoimmune and neurodegenerative diseases. TG2 knock out mice are viable without abnormalities in development and obvious phenotype. But, the analysis of big human genomic databases revealed that the frequency of TG2 protein polymorphism is extremely low and there is no predicted loss of function variant in the human population in homozygous form.
Based on the previous in silico analysis, the PhD candidate will compare the human and mouse TG2 proteins to reveal the role of evolutionally developed 6 new amino acid stretches in humans. This includes the production of chimeric proteins and determination, characterisation of human TG2 interactome in living endothelial cells which very probably influences the physiological endothelial functions.
