Description of the research topic:

The effects of non-steroidal anti-inflammatory drugs (NSAID) on gastrointestinal (GI) mitochondria is controversial, some drugs are inducers of dysfunction (e.g. diclofenac), while others have a protective effect (e.g. acetylsalicylic acid). Among antibiotics, macrolides, tetracyclines, fluoroquinolones and penicillines can also damage mitochondria and the eukaryotic cells by causing the dysfunction of the mitochondrial respiratory chain. Our main question targets the effects of prophylactic and anti-inflammatory GI drugs on epithelial mitochondria, the evolvement of side-effects during their usage, and finally the diagnosis and possible treatment of these side-effects. During our investigations, we aim to analyse human biopsies and mucosal samples from experimental animal model of colitis.
Techniques available: basic laboratory techniques for isolation of epithelial cells from human biopsies. In vivo animal models (rodent colitis), in vivo imaging techniques with fluorescent laser-scanning endomicroscopy or intravital microscopy. Microcirculatory measurements with orthogonal polarization spectral imaging and laser-Doppler technique. Measurement of mitochondrial respiration using high-resolution respirometer and its fluorescent modules (Oroboros O2k).
