Description of the research topic:

The rhizome and roots of this species accumulate many pharmaceutically active substances, belonging to various chemical groups. The most important compounds are the phenolglycoside salidroside and its aglycone tyrosol and phenylpropanoide glycosides of cinnamic alcohol such as rosin, rosavin, rosarin. Since the glycosides are mostly accumulated in the rhizome and roots of the plant, therefore hairy roots should be a reliable alternative for their biosynthesis. Hairy root cultures are initiated by Agrobacterium rhizogenes transformation. These cultures are considered to be genetically and biochemically stable and hence they can be used as a sustainable platform for in vitro biosynthesis of secondary metabolites. In course of the work root and hairy root cultures from different geno(chemo)types of Rhodiola rosea will be established. An efficient method for the initiation and maintenance of the hairy root cultures will be developed. Roots and hairy roots will be analysed for glycoside content, comparing the different genotypes. Biotransformation capacity of the roots and hairy roots will be explored feeding the precursors of the end products, comparing the different genotypes. Expression of the genes earlier identified will be studied in the root and hairy root cultures with or without precursor feeding with real-time PCR.
