The molecular background of therapy resistance of human melanoma

Description of the research topic:

Metastatic melanoma is associated with poor clinical outcome. During the last few years there have been significant advances in the treatment of these deadly disease. The BRAF inhibitor vemurafenib have shown efficacy as monotherapy in patients with previously unresectable or metastatic melanoma with BRAF V600E mutation (approximately 50% of metastatic melanoma are found to have the BRAF V600E mutations). It was one of the first molecularly targeted agents to receive FDA approval for advanced melanoma. Vemurafenib is a potent inhibitor of the kinase domain of the BRAF V600E. It acts through the MAPK pathway, leading to the reduced transcription of genes involved in various cellular responses. Vemurafenib have been shown to improve overall survival in patients with metastatic melanoma compared with chemotherapy. Although responses with BRAF inhibitors are impressive, durability of response is limited due to resistance, and evidence of disease progression can be seen within 6 to 8 months (sometimes even earlier) of starting therapy due to development of resistance mechanism. Unfortunately the molecular mechanism behind the high rate of resistance is still unknown. The major aim of our study is to define the molecular alterations associated with BRAF inhibitor resistance using newly developed drug resistant melanoma models. Techniques will be used include proteome profiler, gene expression, immunhistochemistry and PCR techniques. Different combination of targeted drugs will be applied.
