Description of the research topic:
Coronary Heart Disease (CHD) is the most common cause of death from cardiovascular diseases. It is still unknown what the mechanisms are by which hyperlipidemia interferes with the mechanisms of cardioprotection, thus finding the link between cardiovascular disease and high-fat diet remains an important scope. On the other hand it has been shown that regular recreational physical exercise can reduce many cardiovascular risk factors like elevated triglyceride and low-density lipoprotein cholesterol, and is also associated with various cardiovascular benefits. Previously it has been shown by our research team that voluntary exercise protects the heart of diabetic rats, and this protection is accompanied by the modulation of antioxidant enzymes such as nitric oxide synthase/heme oxygenase system, gasotransmitters.
In the current study, high-fat diet consuming rats will undergo exercise training and inflammatory, antioxidant and inflammasome system (NLRP3) markers will be analysed from heart tissues using molecular biology assays and immunohistochemical analyses. In addition measurements of heart perfusion according to Langendorff and infarct size will be performed.
This study will provide an opportunity of analyzing a novel molecular pathway and mechanism, particularly inflammatory processes (inflammasome activation) in obesity-induced ischemia-reperfusion injury along with its innovative, practical and experimental attenuation.
