Description of the research topic:

Various options can be chosen within the proposed topic. The designed artificial metalloproteins can be suitable for environmental purpose, e.g. water purification or soil regeneration through the overexpression of metal ion-binding proteins within bacteria or suitable host organism. The goal of these studies is to achieve efficient and selective metal ion-binding and accummulation. In parallel, the artificial metallonucleases may be applied for research and therapeutic schemes. The starting point of these studies are fuctional model compounds or natural protein domains as building blocks of allosterically controlled new enzymes. Solid phase peptide synthesis and recombinanat DNA technology are the basic tools for peptide or protein production. The solution structure and interaction of these molecules with metal ions and/or substrates is investigated by pH potentiometry, ITC and by a broad range of spectrometric (UV/Vis, CD, ESR, NMR, Fluorescence, MS) methods.
