Description of the research topic:

The mammalian syndecan family comprises four members: SDC1-4. They belong to the type I. transmembrane, heparan sulfate proteoglycans, although chondroitin sulfate can decorate the extracellular segment. Usually the state-of the-art emphasizes that their main function is being co-receptors. However SDC4 can be consider as a real receptor regulating the activity of Rho-GTPases. During shedding the extracellular domain is cleaved off generating two signals: the extracellular domain with loaded factors/ proteins and the transmembrane – cytoplasmic domains. We focus on the faith of the last one. We unravelled that SDC4 can be endocytosed, and it follows a retrograde transport route. It can escape from the membrane to the cytoplasm and enter the nuclei through importins –RanGTPase pathway carrying cargo that is attached to the ectodomain. SDC4 is enriched in the nuclear speckles. It regulates the cell proliferation, and cytokinesis. Recently its mitochondrial presence is observed. It interacts with the members of the electrontransport system and with some enzime/ proteins of matrix. All together assume a complex process. Our goal is to unravel this step by step. The projects may cover biotechnological issues. We are using plants and fungi as model organisms, as well.
