Description of the research topic:
Maintenance of the redox homeostasis is fundamental for normal metabolic processes and development of plants. The glutathione plays central role in determination of the redox status and in the antioxidant defence. Elements of the antioxidant stress responses will be investigated on tomato plants and Arabidopsis wild type and glutathione peroxidase, glutathione transferase or glutathione reductase insertional mutants after salt- and osmotic stress treatments. The reactive oxygen species will be detected by fluorescent microscope, the enzymatic and non-enzymatic antioxidants by spectrophotometric methods. The alteration of the redox status will be monitored in situ in living cells using the redox-sensitive green fluorescent protein (roGFPs). The relationship between the redox control and growth will be estimated mainly in roots. The expression patterns of glutathione- and growth-related genes will be also detected.
